Mazdutide is a man-made peptide (a small protein-like molecule) built to act like a natural gut hormone called oxyntomodulin. It activates two cell receptors (docking sites on cells) at the same time: the GLP-1 receptor and the glucagon receptor. It is grouped under metabolic and weight-regulation research. It is approved in China for obesity but is not FDA-approved and is considered investigational (still under study and not yet cleared) in most other countries.

Mazdutide is a man-made peptide. A peptide is a short chain of amino acids, which are the building blocks that proteins are made from. It is modeled after a hormone the gut naturally makes called oxyntomodulin. It acts as a dual agonist, meaning it activates two targets at the same time. Those two targets are the GLP-1 receptor and the GCG (glucagon) receptor. It is also known by the research codes IBI362 and LY3305677.

Key point: Mazdutide activates both the GLP-1 and GCG receptors at the same time. Think of it like a key that fits two different locks at once, triggering two separate body pathways: one linked to appetite, and one linked to how the body burns energy.


Research context and categorization

Mazdutide falls under the metabolic and weight-regulation group of research peptides. Metabolism is the process the body uses to turn food into energy. Researchers look at this compound in relation to body weight and blood sugar control. The GLP-1 side of its action is connected to appetite signaling and how the body handles glucose (blood sugar). The glucagon side is connected to energy use and fat stored in the liver.

Beyond obesity and overweight, it has also been studied in the context of type 2 diabetes, a liver condition called metabolic dysfunction-associated steatohepatitis (a buildup of fat and inflammation in the liver), and a form of heart failure. Outside of its approved use in China, all of these areas are still investigational. Any results seen in research settings are not confirmed or approved benefits in countries where the compound has not yet been reviewed.


Status

  • Regulatory status: The National Medical Products Administration (NMPA), which is China's drug regulator, has approved mazdutide. It was first approved for long-term weight management and later for blood sugar control in type 2 diabetes. It is marketed there by Innovent Biologics. It is not FDA-approved (not cleared by the U.S. drug regulator) and not EMA-approved (not cleared by the European drug regulator). In the United States and most other countries, it is treated as investigational.
  • Sport status: Mazdutide is not listed by name on the WADA Prohibited List (the World Anti-Doping Agency's list of banned substances in sport). GLP-1 receptor agonists as a class are currently placed on the WADA Monitoring Program rather than formally prohibited. This status can change, so researchers should always check the current list. The categories to watch are S2 (peptide hormones, growth factors, and related substances) and S4 (hormone and metabolic modulators).

Reconstitution notes (general)

Reconstitution means dissolving a dry powder in liquid to make a working solution. The concentration (how much compound is in each milliliter of liquid) is easy to calculate. Divide the milligrams (mg) of compound in the vial by the milliliters (mL) of bacteriostatic water you add. For example, a 10 mg vial mixed with 1 mL of water gives 10 mg/mL. The same vial mixed with 2 mL of water gives 5 mg/mL.

Dilution and handling notes (compound-specific)

Mazdutide comes as a lyophilized powder (freeze-dried powder) in a vial. Before lab use, it must be mixed with bacteriostatic water. Bacteriostatic water is sterile water with a small amount of benzyl alcohol added to prevent bacterial growth. Standard peptide handling applies. Vials are commonly available in 5 mg, 10 mg, and larger sizes.

Target Concentration

A working concentration near 10 mg/mL is commonly referenced, so the volume of water added is matched to the labeled mass of powder in the vial. Examples include:

  • About 0.5 mL of water for a 5 mg vial
  • 1 mL of water for a 10 mg vial
  • About 3 mL of water for a 30 mg vial

Keeping concentration near 10 mg/mL means the total liquid volume stays small. Using more water to make a lower concentration is also acceptable and can make small measurements easier to read on a syringe.

Mixing Instructions

As a soluble (easily dissolved) GLP-1 class peptide, mazdutide should dissolve without much effort and produce a clear, colorless liquid. To reconstitute the compound:

  1. Aim the water slowly down the inner glass wall of the vial rather than squirting it straight onto the powder cake. This helps the powder dissolve cleanly without clumping.
  2. Let the vial sit and swirl it gently. Do not shake it hard. Vigorous shaking creates foam and puts mechanical stress on the peptide that can damage it.
  3. If any solid material is still visible, swirl gently again and let the vial rest at room temperature for a short time to finish dissolving.

If the solution stays cloudy, looks hazy, has visible floating pieces, or changes color, something is wrong. Do not use a vial in that condition.


Handling and storage

After mixing, store the solution in a refrigerator at 2 to 8 degrees Celsius (about 36 to 46 degrees Fahrenheit) and keep it away from light. A refrigerated solution is generally considered usable for about 4 weeks.

Handling Guidelines

  • Wipe the rubber stopper with an alcohol swab before each draw.
  • Write the mix date on the vial label so you can track how long it has been stored.
  • Discard the vial if the contents turn cloudy or discolored, or if you see floating particles or solid pieces.

  • Treating IBI362 and LY3305677 as different compounds. These research codes point to the same molecule as mazdutide. Logging them as separate reference materials leads to mislabeled vials and confused records.
  • Assuming an approval abroad changes bench handling. Mazdutide is approved in China and is investigational in most other places. That regulatory status does not change how the freeze-dried powder dissolves or how you calculate concentration.
  • Believing it needs forceful mixing. It is a soluble GLP-1 class peptide that should go into a clear, colorless solution with slow, gentle swirling. Hard shaking makes foam and mechanical stress that can damage the peptide, so more force is not better.
  • Guessing at the mass instead of reading the label. Mazdutide is a large molecule (its formula is listed on PubChem), so the labeled milligrams in the vial divided by the water you add is what sets your mg/mL. Eyeballing the powder cake is not a measurement.
