MK-677, also known as Ibutamoren, is a small, man-made molecule that can be taken by mouth. It belongs to a chemical family called spiroindolines. It works as a growth-hormone secretagogue (a compound that signals the body to release more growth hormone) and as a ghrelin mimetic (something that copies the action of ghrelin, a natural hormone linked to hunger and growth). It does this by latching onto the growth-hormone secretagogue receptor (GHSR-1a), the same docking site in the body that ghrelin naturally attaches to.

Research context and categorization

MK-677 is grouped with growth-hormone secretagogues. These fall under the broader category of metabolic and growth-hormone research. Because it is not a peptide (peptides are short chains of amino acids, the building blocks of proteins), it is chemically different from injectable peptide secretagogues, even though researchers often discuss it alongside them.

In research, MK-677 is studied in relation to the growth-hormone and insulin-like growth factor 1 (GH/IGF-1) axis. Think of this axis as a communication line that helps control growth and metabolism in the body. MK-677 is reported to raise circulating levels of GH and IGF-1 by acting through the receptor, not by directly replacing those hormones.

It has been studied in research on body composition (the balance of fat, muscle, and bone in the body), lean mass, and bone mineral density (how solid and strong bones are). Studies have included older adults and people at higher risk of losing bone density. Researchers also discuss it in connection with sleep and appetite because of how it acts at the ghrelin receptor.

  • Regulatory status: Research-only, not FDA-approved. MK-677 reached mid-stage (Phase II) clinical testing for conditions such as growth-hormone deficiency, muscle wasting, and osteoporosis (a condition where bones become weak and brittle), but commercial development was stopped. It has never been approved for any human use and is treated as an investigational compound.
  • Sport status: Prohibited under the WADA (World Anti-Doping Agency) Prohibited List. It is classified within class S2 (Peptide Hormones, Growth Factors, Related Substances and Mimetics), specifically as a growth-hormone secretagogue, and is banned both in-competition and out-of-competition.

Reconstitution notes (general)

For lyophilized (freeze-dried) research powders that are mixed back into liquid, the working concentration equals the milligrams of powder in the vial divided by the milliliters of bacteriostatic water added. Dilution and handling notes (compound-specific)

MK-677 is not a lyophilized peptide, so the standard bacteriostatic-water reconstitution process does not apply to it. It is an orally active small molecule. It typically comes as capsules or a ready-to-use liquid solution, with a common solution concentration of around 25 mg/mL.

Solubility and Solvents

MK-677 does not dissolve well in plain water. Think of it like trying to mix oil and water. It just does not blend easily on its own. That is why finished liquid products are usually prepared in a solvent system rather than water alone. When a lab starts from raw powder, common solvents include DMSO, ethanol, or PEG-400. These liquids dissolve MK-677 far more easily than water does.

If a water-based preparation is tried at higher concentrations, the compound may not fully dissolve. The solution can turn cloudy or leave visible particles at the bottom. Using the right solvent and mixing thoroughly are the practical fixes. Because MK-677 is handled as an oral-form reagent at the lab bench, there are no reconstitution-for-injection steps to follow.


Handling and storage

  • Store finished solutions in the refrigerator at 2 to 8 C and keep them away from direct light.
  • Wipe any bottle or vial closure with an alcohol pad before accessing it, and label the container with the preparation or mix date.
  • As a general guideline, a refrigerated working solution is often used within roughly a four-week window.
  • Discard any material that becomes cloudy, changes color, or shows floating particles or sediment.

  • It is not a peptide, so it does not reconstitute like one. MK-677 is a small spiroindoline molecule, not a short chain of amino acids. Adding bacteriostatic water to it the way you would for a lyophilized peptide vial does not give you a clean working solution.
  • Haze in plain water is not always a bad batch. MK-677 dissolves poorly in water alone. Cloudiness or particles at higher concentrations at the bench often point to the wrong solvent, not a degraded sample. Labs typically use a solvent system (for example DMSO, ethanol, or PEG-400) rather than water.
  • There are no reconstitution-for-injection steps here. This is handled as an orally active reagent at the bench. Treating the vial like an injectable prep adds steps that simply do not apply.
  • "Growth-hormone secretagogue" does not mean the vial contains growth hormone. It is a receptor-acting compound studied in relation to the GH/IGF-1 axis. It does not carry or replace any hormone on its own.
  • Reaching Phase II is not the same as being approved. MK-677 was tested in mid-stage trials, development was stopped, and it was never approved for any use. Being widely discussed in the literature does not change its research-only status.
