Gonadorelin
What it is
Gonadorelin is a synthetic decapeptide whose amino acid sequence is identical to endogenous gonadotropin-releasing hormone (GnRH), placing it in the GnRH / releasing-hormone peptide family. It acts on the pituitary as a releasing factor rather than as a sex hormone itself.
Research context and categorization
Gonadorelin is grouped under the reproductive and endocrine (hypothalamic-pituitary-gonadal axis) category. Because it mirrors natural GnRH, it is studied in the context of pituitary and gonadal signaling, where pulsatile exposure prompts the pituitary to release luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Historically it was used as a diagnostic agent to probe pituitary responsiveness, and pulsatile formulations were investigated for anovulatory infertility linked to GnRH deficiency. In current research and compounding discussions it is commonly discussed in relation to maintaining testicular signaling during exogenous androgen use and to fertility-focused endocrine questions. These uses beyond the withdrawn historical indications are investigational and not confirmed or approved benefits, and outcomes are context dependent.
Status
- Regulatory status: Research-only / not currently FDA-approved and marketed. No gonadorelin product is presently FDA-approved and commercially available in the United States; the earlier branded diagnostic product (Factrel) was withdrawn, and material today generally comes from compounding pharmacies or research suppliers. It historically carried FDA-approved indications for diagnostic pituitary-gonadal testing.
- Sport status: Prohibited under the WADA Prohibited List. Gonadorelin and other GnRH agonists fall under class S2 (peptide hormones, growth factors, related substances and mimetics), where they are prohibited in males because they stimulate testosterone production. In females, GnRH agonists are not prohibited under this provision.
Reconstitution notes (general)
Concentration equals the milligrams of peptide in the vial divided by the millilitres of bacteriostatic water added. For example, a 2 mg vial brought up with 2 mL of bacteriostatic water gives a concentration of 1 mg/mL. Dilution and handling notes (compound-specific)
Gonadorelin ships as a fine white lyophilized (freeze-dried) powder that is reconstituted before use, with common vial sizes around 2 mg, 5 mg, and 10 mg. A typical working range is a 2 mg vial brought up with roughly 1 to 2 mL of bacteriostatic water, landing the concentration between about 1 and 2 mg/mL; larger vials are scaled the same way. The water volume is chosen mainly to land a convenient, easily measured draw volume rather than to respect a solubility ceiling, since gonadorelin is freely water soluble at these strengths.
In practice it dissolves readily and does not typically gel, cloud, or precipitate at ordinary reconstitution concentrations. Add the water slowly against the vial wall, then swirl gently in a slow circular motion rather than shaking, because aggressive agitation can foam the solution and stress the peptide. If a little powder remains, letting the vial rest at room temperature for a few minutes and swirling again usually finishes it. A correctly mixed vial reads as a clear, particle-free solution; persistent cloudiness or visible particulate is a reason to discard rather than use. Gonadorelin is also light-sensitive, so keep preparations protected from light during handling and storage.
Handling and storage
Refrigerate the reconstituted vial at 2 to 8 degrees Celsius and keep it out of direct light. Wipe the rubber stopper with an alcohol swab before each puncture, and label the vial with the mix date. Observe the general four-week (about 28-day) refrigerated window, avoid freeze-thaw cycles, and discard anything that turns cloudy or discolored or that develops floaters.
